SmartAb™ Recombinant Anti-HAVCR2 pH-dependent Antibody (Clone F38-2E2)

The anti-HAVCR2 pH-dependent antibody (clone F38-2E2) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human HAVCR2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for HAVCR2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from HAVCR2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for HAVCR2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human HAVCR2, or Hepatitis A Virus Cellular Receptor 2, also known as TIM-3, is a transmembrane protein that is a negative regulator of T-cell responses. It is an immune checkpoint molecule and a promising therapeutic target for cancer immunotherapy.
