Recombinant Human Antibody clone BAT123, which is specific to HIV-1 gp120. BAT123, which was made against the envelope glycoprotein gp120 of HTLV-IIIB strain of HIV type 1 (HIV-1), is capable of neutralizing HTLV-IIIB in vitro. It also inhibits the fusion between uninfected CD4+ cells and HIV-1-infected cells to form syncytia.

The monoclonal antibody BAT123 was injected intraperitoneally into four groups of mice (six animals per group) at doses of 5 mg/Kg (○), 10mg/Kg (●),20mg/Kg (□) ro mg/Kg (■). The monoclonal antibody PNTU(∆), BAT12(○), or CGP 47 439(□) was injected intraperitoneally into six hu-PBL-SCID mice at dosesof 40 mg/Kg. An hour later, the mice were inoculated intraperitoneally with 10 animal infectious doses of HIV-1 IIIB. Blood was obtained from the tail wein on the days indicated and serum cincentrations of the injected antibody determined by quantitative ELISA. Error bars represent s.e.m.

SCID mice reconstituted with human peripheral blood lymphocytes were inoculated with HIV-I LAb then BAT123 (I mg/kg) CD was given at various times (x axis). Titer of infectious HIV-I recovered from spleen cells at sacrifice 3-4 weeks later is indicated on y axis. Titer of<0.2 TCID/10⁶ splenocytes was considered negative. If either spleen or peritoneal lavage culture was positive, animal was considered infected with HIV-I.

Two weeks after inoculation with 10 MID₅₀ of HIV-I LAI BAT 123 or control antibody (40 mg/kg) was given. Animals were sacrificed at times indicated (before antibody administration in 10, 2 weeks after antibody administration in 20), and titer ofinfectious HIV-I in spleens determined.
