Recombinant human antibody to gp120. 2C6 showed significant binding only to the Gag-Env pseudovirions and not to the gp120 monomer. 2C6 showed low-level neutralization activity against HIV BaL which increased with the addition of sCD4.

Binding specificity of mAb 2C6 and control mAb RV6-26 expressed as Fabs against the following HIV antigens were assayed by ELISA: BaL gp120 monomer, Gag VLPs, and Gag-Env-VLPs (parental strain of Env was BaL).

HIV strain BaL was incubated for 90 min at room temperature with Abs (12.5 µg/ml) in theabsence or presence of 0.075 µg/ml of soluble CD4. Neutralization was detected 48 hours later. HIV immune patient serum was used as a positive control.
