The anti-ICOS pH-dependent antibody (VS-0825-YC160) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human, mouse, and rat ICOS. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for ICOS under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from ICOS at pH 6.0. This pH-sensitive antibody retains its binding affinity for ICOS on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. ICOS, or Inducible T-cell Co-stimulator, is a co-stimulatory receptor expressed on activated T-cells. Its interaction with its ligand, ICOSLG, provides a critical second signal for T-cell activation and is important for T-cell-dependent immune responses.
