The anti-IL13 pH-dependent antibody (clone Tralokinumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL13. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL13 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL13 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL13 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IL13, or Interleukin-13, is a cytokine that is a key driver of allergic inflammation. It is produced by Th2 cells and is involved in the pathogenesis of asthma and other allergic diseases.
