The anti-IL21 pH-dependent antibody (VS-0825-YC186) targets both human and Cynomolgus Interleukin 21 (IL-21) in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL21 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL21 clearance. The antibody operates in a cyclical manner, binding IL-21 in the extracellular fluid and releasing it in acidified endosomes. IL-21, or Interleukin-21, is a cytokine that is a key regulator of the immune response. It plays a role in the differentiation of B-cells and NK cells and is involved in both anti-tumor and anti-viral immune responses.
