The anti-IL22 pH-dependent antibody (VS-0825-YC187) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to IL22 protein. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL22 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL22 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells, while having negligible binding to IL22 expressed on normal cells under physiological pH. IL22, or Interleukin-22, is a cytokine that is a key player in mucosal immunity and tissue repair. It is produced by T-cells and NK cells and acts on epithelial cells to promote tissue healing and protect against infections.
