The anti-IL25 pH-dependent antibody (VS-0825-YC190) targets both human and mouse IL25 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL25 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL25 clearance. The antibody operates in a cyclical manner, binding IL25 in the extracellular fluid and releasing it in acidified endosomes. IL-25, or Interleukin-25, is a cytokine that is a key driver of the Th2 immune response. It is involved in allergic inflammation and host defense against parasitic infections, playing a critical role in the pathogenesis of asthma.
