The anti-IL32 pH-dependent antibody (VS-0825-YC196) targets human Interleukin 32 (IL-32) in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL32 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL32 clearance. The antibody operates in a cyclical manner, binding IL-32 in the extracellular fluid and releasing it in acidified endosomes. Human Interleukin-32, or IL-32, is a pro-inflammatory cytokine that is involved in the innate immune response and is secreted by various cells, including T-cells and NK cells. It promotes the production of other pro-inflammatory cytokines, like TNF-alpha and IL-6.
