SmartAb™ Recombinant Anti-IL34 pH-dependent Antibody (Clone 404.1)

The anti-IL34 pH-dependent antibody (clone 404.1) targets human IL34 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL34 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL34 clearance. The antibody operates in a cyclical manner, binding IL34 in the extracellular fluid and releasing it in acidified endosomes. Human IL-34, or Interleukin-34, is a cytokine that is involved in the proliferation and survival of monocytes and macrophages. It binds to the CSF1 receptor and is a key regulator of the monocyte-macrophage lineage.
