The anti-IL3RA pH-dependent antibody (VS-0825-YC201) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL3RA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL3RA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL3RA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. IL3RA (Interleukin-3 Receptor Subunit Alpha), also known as CD123, is a component of the high-affinity receptor for Interleukin-3. It is crucial for the proliferation and differentiation of hematopoietic stem cells and is frequently overexpressed on leukemic blast cells in acute myeloid leukemia (AML).
