SmartAb™ Recombinant Anti-IL4R pH-dependent Antibody (Clone 6-2)

The anti-IL4R pH-dependent antibody (clone 6-2) targets human IL4R in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL4R acid-switched antibody was engineered by incorporating histidine residues into its variable regions. These histidine substitutions within its Complementarity-Determining Regions (CDRs) enable pH-responsive antigen binding, which leads to enhanced IL4R clearance. The antibody operates in a cyclical manner, binding IL4R in the extracellular fluid and releasing it in acidified endosomes. IL4R (Interleukin-4 Receptor) is a cell-surface receptor that binds to both IL-4 and IL-13. It is a central player in allergic inflammation, and its signaling pathway is a therapeutic target for diseases such as asthma and atopic dermatitis.
