The anti-IL9 pH-dependent antibody (VS-0825-YC224) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL9. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL9 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL9 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. IL-9 (Interleukin-9) is a cytokine that promotes the survival and proliferation of mast cells, T-cells, and B-cells. It is involved in allergic inflammation and host defense against parasitic infections.
