The anti-ITGB1 pH-dependent antibody (VS-0825-YC228) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Integrin β1 subunit (ITGB1). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for ITGB1 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from ITGB1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. ITGB1 (Integrin β1 subunit) pairs with various alpha subunits to form receptors for extracellular matrix proteins. It is crucial for cell adhesion, migration, and survival, playing a key role in normal development and being implicated in cancer progression.
