The anti-ITGB8 pH-dependent antibody (VS-0825-YC230) targets human ITGB8 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-ITGB8 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced ITGB8 clearance. The antibody operates in a cyclical manner, binding ITGB8 in the extracellular fluid and releasing it in acidified endosomes. Human ITGB8 is an integrin subunit that is a receptor for extracellular matrix proteins and a key activator of latent TGF-β.
