The anti-KDR pH-dependent antibody (VS-0825-YC232) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human VEGF Receptor 2 (KDR). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for KDR under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from KDR at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human KDR, also known as VEGFR2, is a receptor tyrosine kinase and the main receptor for vascular endothelial growth factor (VEGF). It is the primary mediator of VEGF-driven angiogenesis and is a major therapeutic target for anti-cancer drugs.
