The anti-KLRC1 pH-dependent antibody (VS-0825-YC234) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human KLRC1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for KLRC1 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from KLRC1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human KLRC1, also known as NKG2A, is an inhibitory receptor expressed on natural killer (NK) cells and T-cells. It binds to HLA-E and helps to downregulate the activity of these immune cells, playing a role in immune tolerance.
