The anti-MAPT pH-dependent antibody (VS-0825-YC248) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human MAPT. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MAPT under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MAPT at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MAPT, which encodes the tau protein, is a microtubule-associated protein that is essential for stabilizing microtubules in neurons. Aggregates of hyperphosphorylated tau, known as neurofibrillary tangles, are a pathological hallmark of Alzheimer's disease and other neurodegenerative diseases.
