Recombinant Mouse Antibody clone 7-10A, which is specific to MHV. MHV-specific mAb 7-10A that is specific for the viral surface glycoprotein and can also neutralize the virus in vitro and protect mice in vivo.

Groups of six mice were immunized ip with different selected phage and then challenged intracerebrally with 10 LD50 of MHV-A59. Phage clones selected by mAb 7-10A: 10.3 (■), 11.1 (●), 11.5 (▲), 12.4 (♦), 12.9 (▼). Clones selected by mAb 5B19: 33.2 (■). Clones selected by mAb 5B170: 9.1 (■), 7.2 (●), 8.3 (▼), 8.1 (♦). Control vector phage, f88.4 (▲). Protection of C57BL/6 mice by clone 9.1 was statistically significant and is representative of two separate experiments.

Two BALB/c mice received a 15-pg injection of purified radioiodinated scFv fragments. At the times indicated, small plasma samples were obtained and circulating levels of scFv fragments were quantitated by densitometric analysis (panel A) of autoradiograms of SDS-PAGE gels (panel B). The estimated half-life was about 6 min.

Microtiter plates were coated with 500 ng/well of viral antigen preparations (■) or uninfected cell lysates (□). The binding of threefold dilutions of purified scFv fragments was detected using 7-10A-specific anti-idiotype antibodies and horseradish peroxidase-labeled goat anti-rabbit IgG antibodies.

The binding of dilutions of mouse or rat hyperimmune sera produced against MHV-A59. mAb 7-10A (Abl). or biotinylated purified rabbit antiviral Ig was determined using peroxidase-labeled species-specific anti-lg or streptavidin.
