  • The study presents a method for preparing antibody-conjugated liposomes (immunoliposomes). Firstly, MBPE is synthesized through a reaction and liposomes are prepared in a nitrogen atmosphere along with other components. Next, aggregates are removed by centrifugation, and the first 4 ml of the liposome suspension is extracted. Then, it is reacted with a specific protein and treated with cysteine to block the maleimide groups. Finally, the liposomes are further purified by density gradient centrifugation, providing a targeted drug delivery system for subsequent experiments.

  • W24V2-11-G is produced by the fusion of mouse hybridoma cells (NS-1 mouse myeloma cells) with spleen cells of C3H/He x BALB/c F- mice, which previously developed a hyperimmune response to MM46 tumor cells. The monoclonal antibody secreted by W24V2-11-G is of IgM type and specifically recognizes a tumor-associated antigen (called MM antigen) distributed on the surface of a variety of breast tumor cells from C3H/He mice. According to the experimental results, the W24V2-11-G IgM antibody was detected in MM46 cells, but not in MM48 cells and normal spleen cells of C3H/He mice. Therefore, W24V2-11-G is a monoclonal IgM antibody against the surface antigen of MM46 tumor cells.
