The anti-MUC1 pH-dependent antibody (VS-0825-YC261) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human MUC1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MUC1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MUC1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MUC1 is a transmembrane mucin glycoprotein that is overexpressed and aberrantly glycosylated in many cancers, including breast, ovarian, and pancreatic cancer. Its aberrant expression and anti-apoptotic signaling make it a key tumor marker and therapeutic target.
