Figure 1 Cell based assays for identification of blocking antibodies to Notch1 and2.

Six sequence unique anti-NRR1 antibodies were identified to efficiently block activation in HEK-Notch1 cells compared to an unrelated control antibody.

ELISA

ELISA experiments were carried out with serial dilutions of antibodies on immobilised NRR domains ofmouse Notch1.

Inhib

Dose response curves were generated by titration of N1_E6 on HEK-Notch1 andantibodies N2_B6 and N2_B9 on HEK-Notch2.

IF

Figure 4 Inhibition of Notch in human NES cells promotes neuronal differentiation.

Figure 4 Inhibition of Notch in human NES cells promotes neuronal differentiation.

NES cells were differentiated for 7 days treated with antibody blockade of Notch1-3. Cells were then stained with the post-mitotic neuronal marker beta-III-tubulin (Tuj1) which showed that inhibition of Notch increased the amount of Tuj1 + cells.
