The anti-NTRK1 pH-dependent antibody (clone CRB0089) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Tyrosine kinase receptor of NGF (TrkA). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for NTRK1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from NTRK1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human NTRK1, also known as TrkA, is a receptor tyrosine kinase that binds to Nerve Growth Factor (NGF). It is essential for the development of the nervous system, and oncogenic fusions of NTRK1 are a driver of several types of cancer.
