Anti-NTRK2 Antibody Prodrug, Protease Activated (VS-1025-YC57)

The Anti-NTRK2 Antibody Prodrug, Protease Activated (VS-1025-YC57) is an antibody prodrug comprised of a recombinant human antibody targeting human NTRK2, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to NTRK2 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to NTRK2 effectively. NTRK2 (Neurotrophic Receptor Tyrosine Kinase 2) is a receptor tyrosine kinase that becomes a powerful, targetable oncogene when its gene is involved in a fusion event, typically resulting from a chromosomal rearrangement. These fusions create a chimeric TRK fusion protein that is constitutively active and drives unchecked cell proliferation across various cancers, including infantile fibrosarcoma and certain brain tumors. The resulting dependence on the constantly active kinase makes these fusion-positive tumors highly susceptible to specific TRK inhibitors.
