The anti-PAF pH-dependent antibody (VS-0825-YC276) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human PAF. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for PAF under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from PAF at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human PAF (Platelet-activating Factor) is a potent phospholipid mediator involved in inflammation, thrombosis, and allergic reactions. It is a key player in diseases like asthma and is a potent activator of platelets.
