SmartAb™ Recombinant Anti-PDCD1LG2 pH-dependent Antibody (Clone CBLFY19)

The anti-PDCD1LG2 pH-dependent antibody (clone CBLFY19) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human PDCD1LG2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for PDCD1LG2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from PDCD1LG2 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human PDCD1LG2, also known as PD-L2, is a ligand for the immune checkpoint receptor PD-1. This interaction is involved in suppressing T-cell responses and maintaining immune tolerance, making it a target for cancer immunotherapy.
