The Anti-RAD51 Antibody Prodrug, Protease Activated (3C10) is an antibody prodrug comprised of a recombinant mouse antibody targeting human RAD51, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to RAD51 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to RAD51 effectively. RAD51 is the central recombinase enzyme of the Homologous Recombination (HR) pathway, the high-fidelity mechanism essential for repairing DNA double-strand breaks and maintaining genomic stability. While essential for normal cell function, tumor cells often become highly dependent on RAD51's activity for survival, especially if they have defects in other repair pathways (e.g., BRCA1/2 mutations). This reliance makes RAD51 and the HR pathway a critical therapeutic target, as its inhibition can lead to catastrophic DNA damage and subsequent tumor cell death.

- The protein encoded by this gene is a member of the RAD51 protein family. RAD51 family members are highly similar to bacterial RecA and Saccharomyces cerevisiae Rad51, and are known to be involved in the homologous recombination and repair of DNA. This protein can interact with the ssDNA-binding protein RPA and RAD52, and it is thought to play roles in homologous pairing and strand transfer of DNA. This protein is also found to interact with BRCA1 and BRCA2, which may be important for the cellular response to DNA damage. BRCA2 is shown to regulate both the intracellular localization and DNA-binding ability of this protein. Loss of these controls following BRCA2 inactivation may be a key event leading to genomic instability and tumorigenesis. Multiple transcript variants encoding different isoforms have been found for this gene.
