The Anti-RAD51B Antibody Prodrug, Protease Activated (1h3/13) is an antibody prodrug comprised of a recombinant mouse antibody targeting human Rad51B, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to RAD51B protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to RAD51B effectively. Rad51B is a critical accessory factor within the high-fidelity Homologous Recombination (HR) pathway, essential for the repair of DNA double-strand breaks and the maintenance of genomic stability. Mutations in the RAD51B gene are classified as tumor suppressor defects and are associated with a moderate, hereditary increase in the risk for developing cancers, notably breast and ovarian cancer. Because of its role in DNA repair, the loss of Rad51B function creates a reliance on other repair pathways, a vulnerability that is exploited by PARP inhibitors in certain tumor settings.

- The protein encoded by this gene is a member of the RAD51 protein family. RAD51 family members are evolutionarily conserved proteins essential for DNA repair by homologous recombination. This protein has been shown to form a stable heterodimer with the family member RAD51C, which further interacts with the other family members, such as RAD51, XRCC2, and XRCC3. Overexpression of this gene was found to cause cell cycle G1 delay and cell apoptosis, which suggested a role of this protein in sensing DNA damage. Rearrangements between this locus and high mobility group AT-hook 2 (HMGA2, GeneID 8091) have been observed in uterine leiomyomata.
