The anti-RANKL pH-dependent antibody (clone IK22-5) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to mouse RANKL. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for RANKL under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from RANKL at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Mouse RANKL (Receptor Activator of Nuclear factor Kappa-B Ligand) is a cytokine of the TNF family that is essential for the formation and activation of osteoclasts. It is a key regulator of bone remodeling, and therapies targeting RANKL are used to treat bone diseases such as osteoporosis.
