The anti-RGMA pH-dependent antibody (VS-0825-YC293) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human RGMA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for RGMA under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from RGMA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human RGMA (Repulsive Guidance Molecule A) is a GPI-anchored protein that acts as a repulsive guidance cue for axons in the nervous system. It is also implicated in inflammation and bone formation.
