The Anti-RNF43 Antibody Prodrug, Protease Activated (VS-1025-YC33) is an antibody prodrug comprised of a recombinant mouse antibody targeting human RNF43, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to RNF43 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to RNF43 effectively. RNF43 is a gene encoding a negative regulator of the Wnt signaling pathway that acts as a tumor suppressor. Inactivating mutations in RNF43 lead to the stabilization of the beta-catenin protein and unchecked Wnt signaling. These mutations are common in pancreatic, ovarian, and colorectal cancers, where they contribute to uncontrolled cell proliferation.

- The protein encoded by this gene is a RING-type E3 ubiquitin ligase and is predicted to contain a transmembrane domain, a protease-associated domain, an ectodomain, and a cytoplasmic RING domain. This protein is thought to negatively regulate Wnt signaling, and expression of this gene results in an increase in ubiquitination of frizzled receptors, an alteration in their subcellular distribution, resulting in reduced surface levels of these receptors. Mutations in this gene have been reported in multiple tumor cells, including colorectal and endometrial cancers. Alternative splicing results in multiple transcript variants encoding different isoforms.
