The anti-SEB pH-dependent antibody (clone 2F6D9G2) targets S. aureus enterotoxin B (SEB) in a pH-dependent manner that affinity is kept high at neutral pH but becomes weaker when pH is lowered, such as in the acidic environment of the endosomes. The anti-SEB acid-switched antibody was engineered by incorporating histidine residues into their variable regions. The introduction of histidine substitutions into its CDRs allowed pH-responsive antigen-binding leading to enhanced SEB clearance. The antibody works in cycles of SEB binding in the extracellular fluid and SEB releasing in acidified endosomes. Staphylococcus aureus enterotoxin B (SEB) is a potent superantigen produced by the bacterium S. aureus that causes food poisoning and can lead to toxic shock syndrome. It triggers a massive, non-specific activation of T-cells, leading to an overproduction of cytokines and a severe inflammatory response.
