SmartAb™ Recombinant Anti-SHH pH-dependent Antibody (VS-0825-YC305)

The anti-SHH pH-dependent antibody (VS-0825-YC305) targets human SHH in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-SHH acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced SHH clearance. The antibody operates in a cyclical manner, binding SHH in the extracellular fluid and releasing it in acidified endosomes. Human SHH (Sonic Hedgehog) is a secreted signaling molecule and a key morphogen that plays a central role in embryonic development. Its dysregulation is linked to various cancers, making it a target for anti-cancer therapies.
