SmartAb™ Recombinant Anti-SLC1A5 pH-dependent Antibody (Clone KM3842)

The anti-SLC1A5 pH-dependent antibody (clone KM3842) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human ASCT2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SLC1A5 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SLC1A5 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human ASCT2, or SLC1A5 (Solute Carrier Family 1 Member 5), is a sodium-dependent neutral amino acid transporter that is overexpressed in many cancers. It plays a critical role in nutrient uptake and metabolism, making it a promising therapeutic target for inhibiting tumor growth.
