Anti-SMARCA4 Antibody Prodrug, Protease Activated (4E5)

The Anti-SMARCA4 Antibody Prodrug, Protease Activated (4E5) is an antibody prodrug comprised of a recombinant human antibody targeting human and mouse SMARCA4, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to SMARCA4 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to SMARCA4 effectively. SMARCA4 is a central component of the SWI/SNF chromatin remodeling complex and functions as a tumor suppressor. Inactivating mutations or deletions in SMARCA4 are found in up to 20% of all cancers, particularly small cell carcinoma of the ovary, hypercalcemic type (SCCOHT), and non-small cell lung cancer. Loss of SMARCA4 function globally alters gene expression, promoting malignancy.

- Introduction
- The protein encoded by this gene is a member of the SWI/SNF family of proteins and is similar to the brahma protein of Drosophila. Members of this family have helicase and ATPase activities and are thought to regulate transcription of certain genes by altering the chromatin structure around those genes. The encoded protein is part of the large ATP-dependent chromatin remodeling complex SNF/SWI, which is required for transcriptional activation of genes normally repressed by chromatin. In addition, this protein can bind BRCA1, as well as regulate the expression of the tumorigenic protein CD44. Mutations in this gene cause rhabdoid tumor predisposition syndrome type 2. Multiple transcript variants encoding different isoforms have been found for this gene.
