The anti-SOST pH-dependent antibody (VS-0825-YC314) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and mouse SOST. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SOST under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SOST at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human SOST, which encodes the protein sclerostin, is a secreted glycoprotein and a key negative regulator of bone formation. It acts by inhibiting the Wnt signaling pathway and is a therapeutic target for osteoporosis.
