The anti-TFPI pH-dependent antibody (VS-0825-YC323) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TFPI. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TFPI under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TFPI at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TFPI (Tissue Factor Pathway Inhibitor) is a protease inhibitor that serves as the primary physiological inhibitor of the extrinsic coagulation pathway. It plays a critical role in regulating blood clotting and preventing excessive thrombosis.
