The Anti-TFRC Antibody Liposome is a novel therapeutic formulation designed to target and deliver bioactive compounds specifically to cells expressing the Transferrin Receptor (TFRC). This liposomal delivery system is engineered to enhance the therapeutic efficacy of various therapeutic agents, including small molecules, proteins, or nucleic acids, by leveraging the overexpression of TFRC in certain cancer cells and other pathological conditions.

- The Tfr antibody were thiolated using a 40:1 molar excess of 2-iminothiolane at room temperature for one hour, following previously described protocols. The resulting thiolated Tfr antibody (OX26-SH) were then combined with PL/pEGFP and PL/pC27 in a weight ratio of peptide to phospholipid of 1:20, and incubated overnight at room temperature. Targeting liposome complexes were subsequently separated using a Sepharose CL-4B column, and eluted with 0.001 M PBS buffer (pH 7.4) from the reaction mixture. The concentration of uncoupled OX26 or Tfr antibody was measured using the NanoOrange Protein Quantitation Kit. The total and uncoupled amounts of Tfr antibody were denoted as C0 and C1, respectively. The coupling efficiency (CE) was calculated using the formula: CE = (1 - C1/C0) × 100%.

- The mouse anti-rat CD71 antibody, clone OX-26, specifically recognizes rat CD71, commonly referred to as the transferrin receptor. This receptor is a homodimeric type II transmembrane protein found on the surface of all proliferating cells and in cells that have a need for iron, such as reticulocytes and capillary endothelium within the brain. Additionally, clone OX-26 exhibits binding to various non-dividing normal tissues.
