The anti-TLR3 pH-dependent antibody (VS-0825-YC331) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human and cynomolgus TLR3. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TLR3 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TLR3 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TLR3 (Toll-like receptor 3) is a pattern recognition receptor that plays a critical role in the innate immune response by recognizing viral double-stranded RNA. Its activation triggers antiviral and inflammatory responses.
