The anti-TNFRSF12A pH-dependent antibody (clone TPP-883) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TWEAK receptor. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF12A under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF12A at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFRSF12A, also known as the TWEAK receptor or Fn14, is a transmembrane protein that is involved in cell proliferation, migration, and angiogenesis. Its overexpression in many cancers makes it a potential therapeutic target.
