The anti-TNFRSF9 pH-dependent antibody (VS-0825-YC341) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human 4-1BB. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF9 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF9 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFRSF9, also known as 4-1BB, is a co-stimulatory receptor expressed on activated T-cells. Its signaling promotes T-cell activation, proliferation, and survival, making it a promising target for cancer immunotherapy.
