The anti-TNFSF18 pH-dependent antibody (clone YGL386) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to mouse TNFSF18. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFSF18 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, mAbs that showed a significant increase in dissociation from TNFSF18 at pH 6.0 were identified. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Mouse TNFSF18, also known as GITRL, is the ligand for the co-stimulatory receptor GITR. It is involved in T-cell activation and is a promising therapeutic target for cancer immunotherapy.
