Anti-TOP1 Antibody Prodrug, Protease Activated (5C2)

The Anti-TOP1 Antibody Prodrug, Protease Activated (5C2) is an antibody prodrug comprised of a recombinant human antibody targeting human Topo1, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to TOP1 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to TOP1 effectively. Topo1 (DNA Topoisomerase I) is an essential nuclear enzyme that relieves torsional stress in DNA during replication and transcription. Highly expressed in most proliferating tumor cells, it is the molecular target for the successful class of chemotherapy drugs known as camptothecin derivatives (e.g., irinotecan). These drugs kill cancer cells by trapping Topo1 on the DNA, leading to lethal DNA breaks.

- This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This enzyme catalyzes the transient breaking and rejoining of a single strand of DNA which allows the strands to pass through one another, thus altering the topology of DNA. This gene is localized to chromosome 20 and has pseudogenes which reside on chromosomes 1 and 22.
