The anti-TSLP pH-dependent antibody (clone MOR04494) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human, cynomolgus, and rhesus Thymic stromal lymphopoietin (TSLP). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TSLP under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TSLP at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. TSLP (Thymic stromal lymphopoietin) is a cytokine that is a key driver of allergic inflammation and promotes Th2 immune responses. It is a central player in the pathogenesis of allergic diseases such as asthma and atopic dermatitis.
