Anti-VTCN1 Antibody Prodrug, Protease Activated (5F6)

The Anti-VTCN1 Antibody Prodrug, Protease Activated (5F6) is an antibody prodrug comprised of a recombinant mouse antibody targeting human VTCN1, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to VTCN1 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to VTCN1 effectively. VTCN1, or B7-H4, is an immune checkpoint ligand that acts as a strong inhibitor of T-cell-mediated immune response. Its frequent overexpression on various solid tumors, including breast and ovarian cancer, contributes to the immunosuppressive tumor microenvironment, allowing the cancer to escape T-cell destruction.

- This gene encodes a protein belonging to the B7 costimulatory protein family. Proteins in this family are present on the surface of antigen-presenting cells and interact with ligand bound to receptors on the surface of T cells. Studies have shown that high levels of the encoded protein has been correlated with tumor progression. A pseudogene of this gene is located on chromosome 20. Multiple transcript variants encoding different isoforms have been found for this gene.
