The anti-VTCN1 pH-dependent antibody (clone 13D12) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human B7-H4. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for VTCN1 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from VTCN1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human VTCN1, also known as B7-H4, is a co-inhibitory immune checkpoint ligand that is frequently overexpressed in various cancers. Its signaling helps suppress The anti-tumor immune response, making it a potential therapeutic target.
