CrossMab (where Mab representative of Monoclonal Antibody) is a novel format of BsAbs derived from independent parental antibodies. This new technique can be used to produce a bispecific antibody (BsAb) with antibody domain exchange based on knobs-into-holes (KIH) method. This “crossover” keeps the antigen-binding affinity and also preserves the two different arms in order to avoid light-chain mispairing. In comparison with natural antibodies, CrossMAbs show higher stability. We have several different CrossMAb designs as in following diagram, such as Fab, VH-VL and CH1-CL exchanged in different regions. CrossMAbFab format exchanges the full VH-CH1 and VL-CL regions; CrossMAbVH-VL format exchanges the VH and VL regions only; CrossMAbCH1-CL format exchanges the CH1 and CL regions of bispecific antibody.
