The DBCO labeling kit includes a linker with a polyglycine sortase attachment site at one end and a single DBCO group at the other. This kit facilitates the use of chemistry for the conjugation of biomolecules that possess an group. When used for antibody conjugation, this kit allows for the attachment of a maximum of two molecules per antibody.

- It is highly advised to purify the labeled antibody away from any excess label and sortase A5 before using it. If the antibody needs to be used immediately, skipping the purification step may be permissible. However, storing unpurified antibody over time may lead to the removal of the label. Any active sortase A5 present will keep cleaving at the recognition sequence, potentially causing the formation of large antibody complexes or complete removal of the poly-Glycine labels, resulting in an unlabeled antibody. Thus, it is strongly recommended to purify the antibody prior to use and storage.

- The sortase A5 enzyme should be stored at -80°C, the reaction buffer at 4°C, the (Gly)5-DBCO label at -20°C, the stop solution AM3 at room temperature, and the purification columns at room temperature.
