What is Elispot?

Elispot is a plate-based immunoassay that enables the detection and enumeration of cytokine- or antibody-secreting cells at the cell level. Principle of the Elispot Assay

The assay is based on a sandwich immunoassay principle, adapted to a membrane surface that captures secreted proteins directly underneath the secreting cells.

Workflow Overview:

1. Plate Coating

Plates are pre-coated with monoclonal capture antibodies specific to the target cytokine (e.g., IFN-γ, IL-2).

2. Sample Incubation

Freshly isolated immune cells (PBMCs, splenocytes, etc.) are stimulated with antigens or mitogens. Activated cells secrete cytokines that are immediately captured by the coated antibodies.

3. Detection Step

After washing, a biotinylated detection antibody binds to the captured cytokine. This is followed by an enzyme (e.g., streptavidin-ALP or HRP) that catalyzes a colorimetric reaction.

4. Spot Development

Upon substrate addition, dark-colored spots form at the sites of secretion, each representing a single cytokine-producing cell.

5. Analysis

Spots are counted manually or using automated Elispot readers. The number of spots correlates directly with the frequency of antigen-specific cells in the sample.
