Summary: Trans-chromosomic mice with engineered chromosomes holding full human immunoglobulin heavy and kappa chain loci, in a mouse immunoglobulin-knockout background, are established. They stably maintain the chromosomes. High-throughput sequencing finds that they well mirror the human immunoglobulin repertoire. Despite slightly altered B cell development and immune response, they have more antigen-specific plasmablast/plasma cell subsets than wild-type mice, enabling efficient hybridoma production.
